Pengaruh Air Lindi Sludge Sampah TPA Sarimukti Terhadap Perkembangan Embrio Praimplantasi Mencit (Mus musculus L.) SW



Penelitian ini dilakukan untuk mengevaluasi pengaruh air lindi sludge terhadap perkembangan embrio praimplantasi mencit (Mus musculus L.) SW. Mencit dengan umur kebuntingan 0 hari dikelompokkan menjadi mencit kelompok perlakuan air lindi sludge (17%, 29%, 50%), mencit kelompok kontrol negatif yang diberi akuabidestilata steril, dan mencit kelompok kontrol positif yang diberi akrilamida (50 mg/kg berat badan) secara intraperitoneal hingga umur kebuntingan 3 hari. Pada umur kebuntingan 3,5 hari, mencit 'dikorbankan' dan kemudian uterusnya diisolasi. Embrio dipanen kemudian diamati di bawah mikroskop. Persentase embrio abnormal pada kelompok air lindi sludge sampah konsentrasi 17%, 29% dan 50% cenderung meningkat tetapi tidak melebihi persentase embrio abnormal kelompok akrilamida. Pemberian air lindi sludge sampah dengan konsentrasi 17%, 29% dan 50% cenderung menurunkan embrio yang mengalami kelambatan perkembangan tetapi meningkatkan malformasi embrio. Berdasarkan hasil penelitian dapat disimpulkan bahwa air lindi sludge TPA Sarimukti mengganggu perkembangan embrio praimplantasi mencit.

The aim of this research is to evaluation municipal sludge waste leachate from TPA Sarimukti to the preimplantation embryonic development. Female mice (8-10 week- old, 30-37 g) with vaginal plug detected was confirmed as day 0 of gestation, and then mice divided in three groups. Leachate-treated mice received three range of municipal sludge waste leachate concentrations (17%, 29%, 50%), negative control group mice. received sterile distilled water, while positive control group mice received acrylamide (50 mg/kg bw). Municipal sludge waste leachate, sterile distilled water and acrylamide were given by intraperitoneal injection until 3 days of gestation. At 3.5 days of gestation, mice were sacrificed and their uterus were removed. The embryos were harvested and examined under a microscope. The percentage of abnormal embryos in the group of waste sludge leachate concentration of 17%, 29% and 50% tended to increase. The percentage of abnormal embryos waste sludge leachate group does not exceed the percentage of abnormal embryos acrylamide groups. Provision of waste sludge leachate with a concentration of 17%, 29% and 50% tended to increases the embryonic malformations. The results of this study indicated that municipal sludge waste leachate from TPA Sarimukti potentially interfere with mice preimplantation embryonic development.
