Pengaruh jumlah sel inang bakteri E.coli BL.21 (DE3) pLysS dan waktu overekspresi pada produksi protein rekombinan firm-C Salmonella typhi
Protein rekombinan Fim-C S.typhi merupakan protein potensial yang dapat dijadikan sebagai kandidat vaksin tifus dan memiliki peluang untuk dijadikan vaksin alternatif dan diproduksi dalam skala besar. Namun, sebelum masuk ke tahap skala besar diperlukan data optimum skala laboratorium mengenai faktorfaktor yang berpengaruh pada proses produksi protein Fim-C Salmonella typhi. Penelitian ini bertujuan untuk mendapatkan informasi mengenai pengaruh faktor jumlah sel inang bakteri E.coli BL21 (DE3) pLysS dan waktu overekspresi pada produksi protein rekombinan Fim-C Salmonella typhi sebagai dasar pengembangan kandidat vaksin tifus. Proses optimasi overekspresi dilakukan dengan menambahkan inducer IPTG (Isopropyl-β-D-thiogalactopyranoside) 0,5 mM ke dalam biakan bakteri sejumlah 2%, 4%, dan 6% dengan waktu overekspresi 4,5, dan 6 jam. Pengukuran konsentrasi sampel ekstrak protein rekombinan Fim-C dilakukan dengan metode BCA (bicinchoninic acid) menggunakan BCA Kit Assay Thermo ScientificTM dan diukur pada panjang gelombang 590 nm menggunakan ELISA reader. Karakterisasi ektrak protein rekombinan Fim-C dilakukan dengan menggunakan metode SDS-PAGE. Hasil karakterisasi SDS-PAGE menunjukkan bahwa jumlah sel inang bakteri E.coli BL21 (DE3) pLysS optimum adalah 4% yang memiliki pita dengan intensitas lebih tinggi pada ±31 kDa dibandingkan dengan jumlah sel inang bakteri E.coli 2% dan 6%, dan waktu overekspresi optimum adalah 4 jam yang memiliki pita dengan intensitas lebih tinggi pada pita ukuran ±31 kDa dibandingkan dengan waktu overekspresi 5 jam dan 6 jam. Hasil penelitian pengaruh jumlah sel inang bakteri E.coli BL21 (DE3) pLysS dan waktu overekspresi pada produksi protein rekombinan Fim-C Salmonella typhi memberikan kesimpulan bahwa jumlah sel bakteri E.coli 4% dan waktu overekspresi 4 jam merupakan kondisi optimum overekspresi protein Fim-C Salmonella typhi.
Recombinant protein Fim-C S.typhi is a potential protein that can be used as a candidate for typhoid vaccine and has the opportunity to be an alternative vaccine and produced on a large scale. However, before entering into a largescale stage, laboratory optimum data on the factors that affect the production process of Fim-C Salmonella typhi protein are required. This study aims to obtain information about the effect of host cell number E.coli BL21 (DE3) pLysS and overexpression time on production of recombinant protein Fim-C Salmonella typhi as the basis for developing candidate of typhoid vaccine. The optimization process of overexpression was done by adding 0,5 mM IPTG (Isopropyl-β-D- thiogalactopyranoside) inducer into bacterial cultures of 2%, 4%, and 6% with 4,5, and 6 hours over expression. The measurement of the concentration of FimC recombinant protein extract samples was done by BCA (bicinchoninic acid) method using BCA Kit Assay Thermo ScientificTM and measured at 590 nm wavelength using ELISA reader. The characterization of Fim-C recombinant protein extract was performed using SDS-PAGE method. The result of characterization of SDS-PAGE showed that the number of bacterial host cell of E.coli BL21 (DE3) pLysS optimum was 4% which had a band with intensity higher at ± 31 kDa compared with host cell number E.coli 2% and 6%, and the optimum overexpression time is 4 hours with a higher intensity band on ± 31 kDa band compared to 5 hours and 6 hours overexpression time. The result of the research of the influence of host cell number of E.coli BL21 (DE3) pLysS and the time of overexpression on recombinant protein production of Fim-C Salmonella typhi concluded that the 4% E.coli bacteria cell count and 4 hours overexpression time is the optimum condition of Fim- C Salmonella typhi.
