Kemampuan senyawa aktif antifungsi dari Khamir asal daun Bintaro (Cerbera manghas L.) dalam mengendalikan kapang Aspergillus spp. pada jagung pakan ternak
Kontaminasi kapang Aspergillus flavus banyak ditemukan pada bahan pangan dan pakan yang berasal dari produk pertanian. Kapang tersebut memiliki kemampuan menghasilkan senyawa metabolit sekunder yang dapat membahayakan kesehatan hewan dan manusia. Senyawa metabolit sekunder yang dihasilkan berupa toksin yang disebut dengan aflatoksin. Penelitian ini bertujuan untuk memperoleh kapang A. flavus dari jagung pakan ternak serta mengetahui potensi senyawa aktif antifungi khamir asal daun bintaro dalam mengendalikan kapang A. flavus. Penelitian ini terdiri dari beberapa tahapan, yaitu isolasi kapang, deteksi aflatoksin pada medium coconut agar, karakterisasi morfologi kapang dan uji konfirmasi pada medium AFPA, uji antagonis, fermentasi khamir, dan uji aktivitas antifungi. Hasil isolasi diperoleh 12 isolat kapang, dengan warna koloni hijau (J1, J2, J3, J4, J5, J6, J7, J9, J10, J12), hitam (J11), dan orange (J8). Terdapat 2 isolat kapang (J1 dan J4) yang menunjukkan pigmentasi orange kekuningan seperti A. flavus dari hasil uji konfirmasi pada medium AFPA. Hasil skrining 18 isolat khamir melalui uji antagonis, diperoleh 6 isolat khamir (T2, T3, T8, T10, T16, T19) mampu menghambat pertumbuhan miselium dan sporulasi dari A. flavus. Tiga isolat khamir potensial (T1, T8, dan T19) hasil skrining uji antagonis dilanjutkan dengan uji senyawa aktif antifungi terhadap dua isolat kapang Aspergillus spp. Ketiga isolat khamir tersebut mampu menghasilkan zona hambat yang berbeda nyata terhadap pertumbuhan miselium kapang. Berdasarkan hasil penelitian diatas, dapat disimpulkan bahwa diperoleh isolat kapang Aspergillus hasil isolasi dari jagung dan senyawa aktif antifungi yang dihasilkan khamir asal daun bintaro mampu menghambat pertumbuhan kapang Aspergillus dari jagung pakan ternak.
Contamination of Aspergillus flavus are commonly found in food and feed ingredients derived from agricultural products. Both molds are capable of producing secondary metabolite compounds that can harm the health of animals and humans. Secondary metabolite compounds produced in the form of toxin called aflatoxin. The aim of this research is to obtain A. flavus from maize and to know ability of bioactive compound from yeast on bintaro leaves (Cerbera Manghas L.) to inhibit the growth of A. flavus. This study consists of several stages, mold isolation, aflatoxin detection in coconut agar medium, morphology characterization of fungi and confirmation test on AFPA medium, antagonistic test, yeast fermentation, and antifungal activity test. The isolation results obtained 12 isolates of mold, with green colony color (J1, J2, J3, J4, J5, J6, J7, J9, J10, J12), black (J11), and orange (J8). There are 2 isolates of fungi (J1 and J4) showing yellowish orange pigmentation such as A. flavus from confirmation test on AFPA medium. Screening of 18 isolates of yeast through antagonistic test, obtained 6 yeast isolates (T2, T3, T8, T10, T16, T19) able to inhibit the growth of mycelium and sporulation of A. flavus. The three potential yeast isolates (T1, T8, and T19) of antagonistic test screening were followed by the antifungal active compound test of two Aspergillus spp. The three isolates of yeast are able to produce zones that differ significantly to the growth of miselium mold. Based on the results of the above research, it can be concluded that obtamed isolates Aspergillus isolated from maize and the antifungal active compound produced by yeast from bintaro leaves was able to inhibit the growth of Aspergillus on maize livestock feed.
